Description:	In this presentation, Drs. Sidransky and Singleton will show how two entirely different research directions have led to complementary advances in the genetics of Parkinson's disease, yielding new insights into disease mechanisms and therapeutic development. Sidransky, who leads the Molecular Neurogenetics Section in the NHGRI Medical Genetics Branch, will share how clinical observations from a Gaucher disease natural history study led to the recognition of the GBA1 gene as a PD risk factor and directed attention to lysosomal pathways in Parkinson pathogenesis. Singleton, an NIH Distinguished Investigator who leads the NIA–NINDS Center for Alzheimer's and Related Dementias, will detail how genetic studies in large kindreds with autosomal dominant PD resulted in the identification of LRRK2 as an important PD gene. Together they will describe the current state of the field of PD genetics, progress in complex genetics, and how global efforts are being made to dramatically expand our understanding of the basis of PD around the world. A key focus will be on how what we have learned about GBA1, LRRK2, and other PD genes is being used to design new therapeutic strategies for this devastating neurodegenerative disorder. The Demystifying Medicine Series, jointly sponsored by FAES and NIH, includes presentations on pathology, diagnosis, and therapy in the context of major disease problems and current research. Primarily directed toward PhD students, clinicians and program managers, this series is designed to help bridge the gap between advances in biology and their application to major human diseases. Each session includes clinical and basic science components presented by NIH staff and invitees. All students, trainees, fellows, and staff are welcome to participate.
