I will discuss our use of high-throughput screening to discover new host factors that are antiviral against arthropod-borne viruses focusing on how viral RNAs are recognized by diverse RNA sensors. Next, I will discuss how the microbiota shapes immune defenses. We will discuss how we can use the simplified Drosophila system to define the functional roles the microbiome plays in altering immunity and how we can use this information to gain insight into how intestinal immunity is established and maintained. As we know the microbiota can directly modulate immunity, but can also indirectly impact immune functions through metabolites and we are elucidating functions of metabolites by screening these small molecules directly for modulatory functions in diverse cell-based assays in human cells. Lastly, I will discuss our implementation of high-throughput screening to discover drugs that are active against individual patient leukemic blasts, and how we are adapting this assay for clinical trials.
