Description:	Neurotrophins such as nerve growth factor (NGF), brain derived growth factor (BDNF) and, glial derived growth factor (GDNF), activate a number of signaling pathways relevant to neuroprotection. However, their poor pharmacological properties and pleiotropic effects resulting from interaction with multiple neurotrophin receptor signaling systems limit their therapeutic application. For these reasons the outcomes of neurotrophin clinical trials have been disappointing. Therefore, a major therapeutic challenge in the field of neurodegeneration has been to create small drug-like molecules that possess strong neuroprotective activity, good pharmacological properties and, high safety and tolerance profiles. Dr. Longo will discuss his pioneering work on the development of non-peptidyl, small molecule, neurotrophin- mimetics that possess optimal medicinal properties, target specific neurotrophin receptors, activate therapeutically relevant signaling pathways and prevent neurodegeneration.
